Research Abstract |
<Material and method> The skin samples of 2-day-old mice and 6-month-old mice (N2 and N6m), and mouse embryos on embryonic days 13 to 18 (E13-E18) were used for observation of anionic sites with light microscopy. The skin samples of N2, E15, and E18 were used for observation with electron microscopy. I Skin of E13-E18, and N2 were incubated with ET,and we observed localization of intraepidermal cleft. II Skin of E13-E18, N2, and N6m were stained with fine cationic cacodylate iron colloid (Fe-cac-f) after the following pre-treatments : a) non-treatment b) hyaluronidase c) heparitinase, chondroitinase ABC,and keratanase respectively, and all of them III Anionic sites within the epidermis of E15 and N2 were neutralized with polyethyleneimine (PEI). IV Skin of E15, E18, and N2 were observed with EM after Fe-cac-f staining. <Results> IET induced suprabasal cleft formation in the epidermis of E14 and E15. . Finally intraepidermal clefts occured only within the granular layr in N2. II-a Anionic sites were found between adjacent cells in the epidermis of E13 and E14. On E15 to E18, anionic sites became abandant throughout all the layrs of the epidermis except the basal layr. As the skin matured, anionic sites concentrated in the granular layr. And anionic sites were also abundant in the intercellular spaces of hair papilla of E16-E18 and N2. II-b Anionic sites in the epidermis did not change so much, but those in the hair papilla were much decreased by hyaluronidase treatment. II-c Anionic sites of both regions were not altered by respective enzyme treatment. Treatment by all enzymes made no change in the epidermis, but decreased anionic sites of the hair papilla. III PEI perfusion resulted in intraepidermal splitting that occured within the suprabasal layr in the epidermis of E15, and within the granular layr in the epidermis of N2. IV We observed that Fe colloidal particles were distributed diffusely in intra- and intercellular spaces of any cells of all layrs in the epidermis of mice. Specific anionic sites were not found in this study. But specific anionic sites were found in intercellular spaces of hair pappila. Less
