Research Abstract |
Correlation between the number of apoptotic cells and expression of the apoptosis-related antigens (bcl-2, p53 and c-myc) during histogenesis of human tongue cancer was studyed. We collected samples from hyper keratosis (ED), epithelial dysplasia (ED) and squamous cell carcinoma (SCC) from biopsy and the tongue from autopsy cases. The tissues of these lesions were fixed in neutral phosphate buffered formalin and embedded in paraffin. Serial sections cut at 4mu m were mounted on silane-coated glass slide. Apoptotic cells were determined by the nick end-labeling method (TUNEL) and immunohistochemically staining for Le^Y. The deparaffinized sections were immunostained by streptoavidin-biotin complex methods using Pathostain ABC kit. The numbers of positive cells were counted by using a computer-accessed image analyzer (IBAS 2000), and then calculated Apeorance Index (AI) and Labeling Index (LI). These lesions were compared with normal mucosa adjacent to them. Autopsy materials were not stained immunohistochemically, but we obtained good staining results in HK,ED and SCC from biopsy samples. In each lesion, the AI and LI by TUNEL method were 18.0 and 1.3 in normal mucosa, 23.3 and 2.2 in HK,25.8 and 3.3 in ED,and 50.2 and 3.9 in SCC,respectively. And, the AI and LI by immunohistochemically staining for Le^Y were 15.4 and 0.8 in normal mucosa, 16.2 and 1.2 in HK,19.2 and 1.8 in ED,36.6 and 2.7 in SCC,respectively. However, the AI and LI of bcl-2 positive cells were 42.1 and 38.2 in normal mucosa, 38.0 and 26.2 in HK,12.0 and 2.3 in ED and 7.6 and 1.9 in SCC,respectively. The AI and LI of p53 positive cells were significantly higer in ED and SCC,and the AI and LI of PCNA positive cells were significantly higher in in HK,ED and SCC. These results suggest that p53 (stimulative) and bcl-2 (suppressive) may regulate apoptosis during the histogenesis of the tongue cancer.
