| Research Abstract |
マウス皮膚損傷の治癒過程における,炎症性サイトカインインターロイキン1β(IL-1β)の動態について,蛋白質及びmRNAレベルで解析した.
材料:マウス背部の皮膚に陳旧度の異なる切創を作成し,損傷部位を経時的に採取して試料とした.IL-1β及びIL-1βmRNAの定量的検討:採取した損傷部位をホモゲナイズし,遠心した後,その上清に含まれるIL-1βをELISA方で定量した.IL-1βは受傷後3日目で再び上昇に転じ,いわゆる"リバウンド"現象が観察された.IL-1βmRNAについては,採取した損傷部位から組織内mRNAを抽出し,RT-PCR(逆転写酵素-ポリメラーゼ連鎖反応)法によりIL-1βmRNAのみを特異的に増幅し,増幅試料を電気泳動したところ,受傷後3時間および1日目の損傷部位でIL-1βmRNA由来のバンドが検出された.
組織切片上でのIL-1βmRNA局在様式の検討:上述のRT-PCR法の結果に基づき受傷後1日目の損傷について,採取した損傷部位をアセトンで固定後,凍結切片を作成し,in situ RT-PCR法によりIL-1βmRNAの局在を検討したところ,創縁付近に遊走してきたマクロファージの核に陽性所見が観察された.しかしながら,検体によってシグナルの強度が不安定となる傾向にあるため,最良のシグナルを得るための条件について,種々検討中である.至適条件が決定された後,異なる受傷後経過時間の損傷についてもIL-1βmRNAの局在を検索する.
まとめ:IL-1β及びIL-1βmRNAの定量的検討結果は,早期炎症反応のケミカルメディエーターであるIL-1βが,損傷局所において経時的に新しく産生されていることを実証している.したがって,このIL-1βmRNAを指標として,遺伝子レベルで生活反応の有無や受傷後経過時間を判定することも十分可能であると考えられる.平成9年度では損傷治癒過程におけるIL-1β産生細胞の経時的変化を中心に,検索対象として新たにTNF(腫瘍壊死因子)-αを追加して研究を進めていく予定である.
