| Research Abstract |
昨年までの研究で、水晶振動子を用いて結合・解離反応を行わせ、その時の振動数変化を測定することにより、ビオチンの質量測定することを試みた。水晶振動子にリオ酸修飾ベータガラクトシダーゼを吸着させ、アビジンを加えたときに振動数は減衰した。これを基にし、今年度は、抗原抗体反応にもとづく免疫センサーの開発を行っており、光原抗体反応後は振動数の減弱を確認している。また、新しい概日リズム時計を構成する蛋白質を昨年3つクローニングした。最近、その配列を基にして、合成ペプチドを作り、それに対する抗体を作成した。現在、この抗体の特異性を免疫組織化学、ウェスターンブロッティング法にて確認し、その抗体を利用して、時間遺伝子のリアルタイムモニタリング系を構築中である。来年度は、最近開発された安定して連続測定の可能なエバネセント波蛍光免疫測定法を用いて、視交叉上核培養系から放出されたペプチド量を測定すること、バイオルミネッセンスによる連続測定を計画中である。これは、高感度フォtン検出装置を用いて、視交叉上核培養系の発する生体微弱光(バイオルミネッセンス)を高感度フォトン検出装置にて検出する予定である。
