Research Abstract |
A traumatic spinal cord injury model was established using rats. A 5Ogr.closing pressure clip was used for this spinal cord clip injury model. An arterial diameter at the dorrsal surface of the thoracic spinal cord was measured using vital videocamera system and tissue blood flow of the spinal cord was measured by laser Doppler flowmeter. A quantitative study of total Evans-blue fluorescene was performed at 6 hours after clip injury in an atempt to analyze the severity of the spinal cord-vessel barrier by measuring the leakage of fluorescene. The amount of microcirculatory disturbance was estimated by measuring the extent of decreased carbon filling area in the spinal cord. The aim of this study was to investigate the effect of the administration of the anti-CD agents for the possible improvement of the microcirculatory disturbance which might result in preferable outcome of neurological status and histopathological point of view. As a result, major histological change was devided into two categories, hematoma and contusion. More prominent disturbance of microcirculation was recognized in the rostral part to the injury site than the caudal part. Among three groups, sham group(3 animals), vehicle group(6 animals) and anti-CD group (6 animals), microcirculation was preserved in best grade in anti-CD group, followed by vehicle group, sham group, respectively. The size of the spinal cord hematoma was smallest in anti-CD group, and largest in sham group. A spinal cord - vessel barrier was best preserved in the anti-CD group by means of measuring the total volume of Evans-blue fluorescene. This result was statistically significant in comparing the other two groups. From the histological point of view, the best result was achieved in the anti-CD group than in the sham and vehicle groups. In summary, anti-CD group obtained good results in terms of both histology and neurology presumably due to inhibitory effect of the circulating neutrophil suppression in the microcirculation.
