覚せい剤による依存形成に関与する新規機能分子の同定と生理機能の解明

Research Abstract

覚せい剤や麻薬による薬物依存は大きな社会問題であるが,治療法がほとんど確立されていないのが現状である.治療薬の開発が遅れている理由として,依存形成の鍵となるタンパクが明らかになっていないことが挙げられる.そこで,PCR-select cDNAサブストラクション法を用いて,薬物依存形成の鍵となる新規機能分子の探索を試みた.その結果,覚せい剤メタンフェタミンを連続投与されたマウス脳側坐核から新規機能分子'shati'を同定した.昨年度までは、shatiが,メタンフェタミン誘発自発運動量亢進および場所嗜好性の形成に対して抑制的に働くことを明らかにした.さらに,shatiがメタンフェタミンによって引き起こされる細胞外ドパミン量の増加およびドパミン再取り込み能低下を抑制することを示した.本年度は,shatiのドパミン再取り込みにおける機能の詳細なメカニズムを明らかにするために,shatiの発現ベクターを構築し,shatiを過剰発現させたPC12細胞を用いて,shatiの生理機能の検討を行った.
PC12細胞へshati発現ベクターをトランスフェクションすると,コントロール細胞と比較して,shati mRNA発現量の増加,ドパミン再取り込み能の促進,メタンフェタミン誘発ドパミン再取り込み能低下の抑制が観察された.
メタンフェタミンはドパミントランスポーターに作用して,ドパミンの再取り込みを阻害する一方で,シナプス間隙のドパミン遊離を促進する.その結果,自発運動量亢進および場所嗜好性の形成が引き起こされると考えられる.本研究の結果より,shatiは,ドパミン再取り込みを促進し,ドパミン遊離を抑制することにより,シナプス間隙のドパミン量を減少させ,メタンフェタミン依存形成を抑制していることが示唆される.
今後,shatiの依存形成メカニズムへの関与をさらに詳細に解明することが,薬物依存形成機構の解明と治療薬の開発に繋がると考えられる.
