Research Abstract

1.ショウジョウバエの神経系で発現するカドヘリンを2種類同定し、その解析を進めた。その一つDN-カドヘリンの変異体を調べた結果、中枢神経系における軸索の走行パターンに種々の乱れが認められ、その過程における必須の役割が証明された。もう一方の分子は以外にも7回膜貫通ドメインを有し、その変異個体では中枢神経系の繊維束に断絶が生じており、シグナル伝達に関わる重要な役割が示唆された。また、昨年度に得られたカドヘリン-11のノックアウトマウスの解析の結果、脳の局所的な異常が認められた。
2.新たな神経-筋認識候補分子として、leucine-rich repeat遺伝子ファミリーに属する膜貫通型蛋白質P750をクローニングした。この分子は神経-筋結合の特異性に関連し、特定の運動神経細胞、筋肉において発現することから、神経の遊走、標的認識に関与している可能性が高いと判断された。
3.網膜分化を誘導するHLH型因子としてATH-3とMashlが、抑制因子としてHES1が同定された。また、これら以外にもHLH型因子であるHES5が網膜に発現していた。したがって、網膜分化は少なくとも4種類のHLH型因子によって誘導あるいは抑制されており、これらの因子がバランス良く機能することによって眼の形態形成が進められることが示された。
4.予定細胞死領域となっている第3指間から指間間充織を取り出し、培養系でのBMPの作用を解析した。この培養系に様々なTGFβファミリー蛋白質を添加すると、アクチビンやTGFβでは細胞数に変化は起きないが、BMP-2,BMP-4では細胞死が引き起された。指間間充織細胞に、あらかじめドミナントネガティブレセプターをウィルスベクターを用いて導入しておくと、BMP-2,BMP-4に対して抵抗性を示した。これらの事実はBMP-2,BMP-4がプログラム細胞死の特異的メディエーターとなっていることを強く支持する。
5.転写因子Islet-3のLIM領域のみをゼブラフィッシュ胚で過剰発現させ、内因性Islet-3の機能を抑制する実験を行った。その結果、眼胞の欠失、視蓋及び小脳への区分化の阻害、中脳・後脳境界部での細胞死による小脳原基の低形成がおきた。さらに、LIM過剰発現胚では、中脳・後脳境界部におけるWntlの発現が消失し、その後にEn2とPax2の発現も失われることが分かった。これらの結果から、中脳及び中脳・後脳境界領域の分化においてIslet-3は、Wntlを分泌する過程の維持に必須であることなどが示唆された。
6.神経系特異的RNA結合性蛋白質をコードするmouse-musashi-1に関してノックアウトマウス作製に成功した。詳細な表現型の解析を通して、本遺伝子産物の哺乳類中枢神経系幹細胞の非対称的分裂における役割を検討中である。
7.エピブラストから始原生殖細胞が誘導される分子機構を明らかにする目的で、エピブラスト内の予定始原生殖細胞領域で発現する遺伝子のクローニングをめざし、EG細胞-ES細胞のサブトラクションクローニングを試みた。その結果、EG細胞で特異的に発現する16種類の遺伝子断片を得ることに成功した。さらに、原腸陥入前の胚から単離したエピブラストから始原生殖細胞が誘導される初代培養系を確立することに成功した。
以上の他にも、多くの業績をあげることができた。
