Research Abstract |
【目的】 エネルギー代謝調節系は摂食調節系とエネルギー消費調節系より成る。レプチンは摂食調節系の主要な分子だが、本研究ではレプチンの下流に位置し、エネルギー消費調節に関与する脱共役蛋白質3(UCP3)の遺伝子発現調節機構を検討した。エネルギー代謝の状態を伝達する核内転写因子としてPeroxisome Proliferator-Activated Receptor(PPAR)が注目されているが、本研究ではPPARγアゴニストで、インスリン感受性増強薬のチアゾリジンによるUCP3遺伝子発現調節を検討した。 [方法] 遺伝性肥満Wistar fatty ラットでチアゾリジンのピオグリタゾン3mg/kg/日2週間投与し、UCPファミリー遺伝子発現を検討した。SDラット副精巣周囲白色脂肪初代培養でピオグリタゾンによるUCP3遺伝子発現調節を検討した。 [結果] ピオグリタゾン非投与では、Wistar fattyラットのUCP3遺伝子発現はWistar leanラットに比べて後腹膜脂肪、褐色脂肪、骨格筋で有意に38%、45%、76 %低下していた。ピオグリタゾン投与Wistar fattyラットでUCP3遺伝子発現は副精巣周囲白色脂肪、後腹膜脂肪、褐色脂肪で2.1倍、2.0倍、1.6倍に有意に上昇した。ラット白色脂肪細胞初代培養系でUCP3遺伝子発現はピオグリタゾン10-5M以上の濃度で、用量依存性に有意に上昇した。 [考察] Wistar fattyラットでのUCP3遺伝子発現の低下の機序としては、レプチン系の異常によるものと考えられた。チアゾリジンは褐色脂肪、白色脂肪でUCP3遺伝子発現を増加させることが明らかとなった。
