Research Abstract

Ki-67 was immunohistochemically investigated in paraffin-embedded preparations according to the performance report for the previous year. Needle biopsy specimens were obtained from 106 untreated patients with prostatic cancer. At least 500 cancer cells were evaluated in a tissue preparation. The Ki-67- positivity rate was calculated and used as the Ki-67 labeling index. The above examination was performed by one physician. Less than 500 cancer cells were evaluated in 118 of 106 patients, thus these cases were excinded from calculation of the mean Ki-67 labeling index. The mean Ki-67 labeling index of 88 cases was 13.6% and the median value was 8.1%. Regarding the relationship between the Ki-67 labeling index and the degree of tumor- malignancy, the Ki-67 labeling index was higher in groups with a higher Gleason score, showing that the Ki-67 labeling index highly correlated with the degree of malignancy. As a result, the survival period was significantly shorter in the group with 8.1% or higher Ki-67 labeling index. Based on these findings, it was confirmed that the Ki-67 labeling index reflects the degree of malignancy and prognosis in prostatic cancer, and thus, in an appropriate parameter of prostatic cancer growth activity. In contrast, estabhishrnent of appropriate conditions for immunohistochemical staining of vascular endothelial growth factor was not successful and is under investigation. Regarding immunohistochemical investigation of the protein encoded by KAI 1 metastasis suppresser gene and androgen receplor gene analysis using frozen samples, since the number of frozen samples was not sufficient for either procedure, appropriate conditions could not be established, and the conditions for both procedures are under investigation, including additional sample collection. ln consequence, the relation of prostatic cancer growth activity with either vascular endothelial growth factor or KAI 1 metastasis suppresser gene expression could not be investigated in this study.
