Research Abstract

To clarify the mechanism involving placental development and functions, we have cloned a new placenta-specific kinase gene from a placental cDNA library and named Polo like kinase(PLK). The Northern blot analysis demonstrated the highest expression only in the placenta but not in other tissues including the brain, lung, liver and kidney [Hamanaka et al. Cell Growth & Differ. 5 : 249-257, 1994].
The immunohistochemical study of the placenta showed that human PLK was strongly expressed in the syncytiotrophoblast during the early stages of pregnancy [Yoshimatsu et al. Res.Commun.Mol.Pathol.Pharmacol. 106 : 3-12, 1999]. Murine PLK was strongly expressed in troploblastic tissue invading the decidua and in trophoblastic tissue around embryos [Takai et al. Reprod.Fertil.Dev. 11 : 31-35, 1999]. In normal human endometrium during the menstrual cycle, PLK is associated with estrogen-dependent cellular proliferation and estrogen functions may be involved in its regulation [Takai et al. Reprod.Fertil.Dev. 12 : 59-67, 2000]. Furthermore, PLK expression in ovarian cancer and endometrial cancer is associated with high histological grade [Takai et al. Cancer Lett. 164 : 41-49, 2001].
