Research Abstract

A CMP-NANA containing fluorescence probe as an energy donor and an alkylactosaminide containing fluorescence probe as an energy acceptor was successfully synthesizedegio- and stereoselectively as substrates for sialyl transferase. These substrates showed clear emission spectrum in which emission peaks were at around 340 and 525 nm due to 2-naphthyl and dansyl emissions, respectively, when the substrates was excited at 280nm. The synthetic substrates were used as substrates for SiaT based on fluorescence energy transfer. Vmax and Km were determined as 0.119 mol/min and 0.85 M, respectively. These substrates provide tremendous advantages such as sensitivity, speed, and feasibility of continuous assay.
