Research Abstract

ゴナドトロピンレセプター及びTSHレセプターcDNAが明らかとされて以来、その構造と機能の関係が明らかとされてきている。これに伴い、各々の分野でのレセプター異常と病態との関連も次々と明らかとなっている。特に、レセプターがホルモンの非存在化に活性化される思春期早発症などが良い例である。両研究者の共同により、レセプター機能の調節について更なる検討を加えることを目的とした。
ゴナドトロピンレセプターは、TSHレセプター同様、7回膜貫通のG蛋白結合型レセプターである。これらのレセプターとホルモンとの間に特異性が保たれているのが一般的であるが、一定の病態ではこの特異性が低下することが考えられる。先天性甲状腺機能亢進症ではTSH高値になり、このTSHが性腺のFSH/LHレセプターに反応し、思春期早発症を誘引することが知られている。さらに、家族性甲状腺機能亢進症(原因がTSHレセプターの構造異常がある場合)においては、妊娠中のhCGに反応して、妊娠中の甲状腺機能亢進症が特に悪化することが最近証明された。さらに我々は、ヨーロッパ人と日本人との間でのLHレセプター遺伝子に相違のあることを発見した。しかし、アミノ酸で2個分の相違は機能的には変化を起こさないことも判明した。報告されてきたレセプター構造上の変異は、それがただ1つのアミノ酸の変異であっても、機能に影響を及ぼすことが多かったので、今回の結果は興味深いものであり、今後さらにレセプターの構造と機能の関係の詳細な解明が必要と考えられる。
