| Research Abstract |
炎症性生理活性脂質の機能や、アレルギー、ショックなどの病態における役割を解明し、最終的には受容体の発現やその作用を制御する方法、薬物を開発する目的で、以下の研究を行った。
1.PAF受容体の機能の解明
すでに確立しているPAF受容体トランスジェニックマウス、ノックアウトマウスを用いて、正常マウスとの比較を、塩酸注入及びLPS投与の急性肺炎症モデルにおいて行いPAF受容体の果たす役割を個体レベルで検討した。その一部を論文にまとめ発表した。
2.LTB4受容体の遺伝子発現制御と機能の解析
(1)ヒトLTB4受容体のゲノムの解析とプロモーター機能
ゲノム遺伝子の解析により、ヒトLTB4受容体のプロモーター領域の機能をレポーター遺伝子を用いたアッセイにより明らかにした。
(2)マウス、ラット、モルモットLTB4受容体遺伝子のクローニング
ノックアウトマウス作製のためのクローニングとターゲティングベクターの作製を行った。また、ラットにおいて炎症性刺激による受容体遺伝子の誘導を報告した。モルモット遺伝子のクローニングにも成功し、ヒト遺伝子との機能比較を行った。
