Research Abstract

Due to the development of molecular biology techniques, several types of neurotrophic factor secreting cells can be created. Glial cell line-derived neurotrophic factor (GDNF) is one of the most potent neurotrophic factors for dopaminergic neurons. In order to analyze the efficacy of GDNF secreting cell grafting, encapsulated GDNF secreting cells were grafted into the striatum of hemiparkinsonian model rats.
We have established a cell line which produces GDNF in a stable fashion by introducing GDNF gene into the baby hamster kidney (BHK) cells using liposome method. Parkinsonian model rats were created by unilateral injection of 6-hydroxydopamine (6-OHDA) into the striatum.
Animals receiving GDNF capsule showed significant recovery of nigrostriatal dopaminergic system morphologically and behaviorally. Chemical analysis of retrieved caspules revealed that the capsules continued to secrete GDNF at least up to 6 months after transplantation. Histological analysis of the capsules showed that GDNF secreting cells survived well in the capsule for a long period of time.
These results support the idea of using neurotrophic factor secreting cells for the treatment of Parkinson's disease. Since neurotrophic factor delivery is considered effective for several types of neurological disorders, the technique of encapsulated cell grafting can also be applied to other neurological disorders.
