インヒビンダイマーとαモノマーの局所作用

Research Abstract

下垂体前葉初代培養細胞系で、インヒビンダイマーがFSHの分泌を抑制するのに対して、インヒビンαモノマーはFSH分泌抑制活性を示さないことが報告されている。しかし、インヒビンダイマーについてはこれまでの報告で作用の見られる組織と見られない組織があることより、インヒビンαモノマーについても組織や細胞により感受性が大きく異なる可能性が考えられる。このため、局所作用を観察するに適した系を確立する目的でインヒビンαサブユニットの発現が報告されている種々の細胞について検討した。インヒビンダイマー、インヒビンαモノマーの量が限定されているため、まず、リコンビナントアクチビン・Aを用いて検討した。 1)下垂体細胞:ラットの初代培養下垂体細胞からのFSH分泌についてはインヒビンαモノマーがFSH分泌抑制活性を持たないことは既知なため、インヒビンαサブユニットとインヒビン/アクチビンβサブユニットの発現が報告されているラクトトローフ系の細胞株について検討し、プロラクチン産生がアクチビンにより制御されていることを確認した。 2)卵巣細胞:DES処理幼若ラットの顆粒膜細胞とエストロゲン未処理幼若マウスのoocyte-granulosa cell-basement membrane-theca cell complexがアクチビンに鋭敏に反応することを確認した。 3)胎盤:ヒト胎盤は卵巣と並ぶインヒビン産生器官であり、合胞体栄養細胞にインヒビンαサブユニットの発現が報告されているため、満期胎盤の培養細胞について検討し、アクチビンのhCG産生促進能を指標とする限りは、この細胞は目的には適さないことが明らかとなった。
