抗ゴナドトロピンレセプター抗体の検討

Research Abstract

ゴナドトロピンに対する抗体については、今までこれを解析する手段がなかったが、レセプターをCHO cellに導入し恒常的レセプター発現cell lineを作ることにより多数の検体を処理する事が可能になり、この病態を解析する事の現実性が出てきた。THSレセプター抗体の中には、刺激型と阻害型の抗体が存在するので、卵巣や精巣へのレセプター抗体の存在の解明を目的とした。
刺激型の抗体に関しては、Basedow病の患者でTHSレセプター抗体の存在する血清によるLH/hCGレセプターに対する作用を検討した。この実験により、Basedow病患者における排卵機能障害の機序について、甲状腺ホルモンの生殖腺細胞に対する影響以外の新しい病能が明らかとなる可能性がある。
TSHレセプター抗体を保持する患者の血清を用いて、FSHレセプターを発現させたCHO細胞に作用させたところ、有意な作用を示さなかった。このことは、TSHそのものがFSHレセプターを認識する可能性は存在するものの、TSHレセプター抗体がFSHレセプターを認識する可能性が少ないことを示す結果である。
今後はさらに、レセプターの機能上抗原性の高い部分のアミノ酸配列を含むペプチドを合成して抗体を作製し、その機能についても検討する予定である。
