Research Abstract	本年度は、加齢現象にともなう骨の形態変化あるいは分子細胞学的変化を検索し、骨組織の形態制御機構解明の一助とすることを目的として、(A)電子エネルギー損失分光電子顕微鏡とイメージングプレートとを組み合わせた分析電顕システムを用いた石灰化基質形成機構の解析、(B)加齢現象に伴う関節形態の変化に関する検索を行った。 (A)に関しては、急速凍結・凍結置換した胎生期ラット頭頂骨を観察した。その結果、石灰化していない類骨においては、コラーゲン細繊維の間隙にプロテオグリカンの網目構造が観察され、Caはプロテオグリカンの領域に、Pはコラーゲン細繊維の領域に、相対的に集積していることが明かとなり、それぞれの構造がCaおよびPを保持し、カルシウム・リン酸イオン積の上昇を抑制する事により、未石灰化状態を維持しているものと思われた。一方、石灰化が進行する石灰化球ではCaとPの両者が共在化しており、Ca・Pの共在化機構が石灰化の進行に重要な役割を果たしていることが明らかとなった (B)に関しては、ラット脛骨近位部を用いて関節軟骨の経時的変化を形態学的に追跡したところ、生後2ヵ月から8ヵ月にかけ、関節軟骨の幅が減少し、関節軟骨の下層に軟骨下骨が添加されて血管進入部位と肥大化軟骨細胞の隔絶が生じ、成熟した関節軟骨へと変化する過程が明らかとなった。
