| Research Abstract |
LPS不応性のヒト肺胞上皮細胞(A549cell)におけるIL-8産生誘導能を指標として、緑膿菌の培養上清(CSPA)から分泌型IL-8誘導因子の精製をすすめた。まず、CSPAをイオン交換クロマトグラフィーにアプライするために、20mMTris/HClで透析した(d-CSPA)。このd-CSPAのIL-8誘導活性はCSPAに比較して大差なかった。このd-CSPAをQカラムにアプライし、1M NaClのグラジエントで溶出したところ、グラジエント16-20%のところで単一のIL-8誘導活性を検出した。さらに、透析後にMono-Qカラムにアプライし、1M NaClのグラジエントで溶出した。やはり、グラジエント16-20%付近で単一のシャープなIL-8誘導活性が検出された。今後、さらに生化学的同定をすすめ、A549cellにおけるIL-8遺伝子誘導における転写調整因子について解析する。
