成人T細胞白血病の多段階発癌に関わる遺伝子の同定と解析

Research Abstract	1)成人T細胞白血病(adult T-cell leukemia:ATL)細胞におけるメチル化の異常を明らかにするためにDNA methyltransferase遺伝子の発現を解析したところ細胞株、新鮮白血病細胞においてDnmt1の発現抑制が認められた。一方、de novo methylationに関連するDnmt3に関しては発現抑制は認められず、ATLにおける発がん機構においてDnmt1発現抑制がメチル化の異常、ひいては低メチル化による発現増強、ゲノム不安定化をもたらしているものと考えられた。今後は、Dnmt1発現抑制機構の解明、メチル化異常の標的遺伝子の単離を行っていく予定である。 2)HTLV-I感染者における免疫不全状態の機序を明らかにするために解析を行い、HTLV-I感染者では特異的にナイーブTリンパ球の減少が見られ、一方、メモリーTリンパ球は増加していることを明らかにした。胸腺機能の指標であるT-cell receptor gene excision circle(TREC)をsequence detectorを用いて定量したところHTLV-I感染者では胸腺レベルにおけるTリンパ球の産生障害が認められることが明らかとなった。このようなナイーブTリンパ球の減少による免疫不全の結果としてHTLV-I感染者ではEpsten-Barr VirusのDNA量が増加していることを示した。このようなHTLV-I感染による免疫不全はATLの発症にも関連していると考えられる。
