Research Abstract	糖鎖の機能を正確に知るためには、複雑な構造の糖質がつながった糖鎖やその誘導体を人工的に合成して、その性質や生物学的機能などを評価する必要がある。糖鎖を人工的につくるには、これまで主として触媒や有機溶媒を用いる化学合成法が有力な方法であったが、近年、より安全で環境にやさしい方法として酵素を用いる糖鎖合成の研究が盛んになっている。いずれの方法を用いるにしても、「糖鎖の自動合成」は未解決の大きな課題で、多くの化学者がさまざまなアプローチでこの難題に挑戦しているのが現状である。酵素の反応を基軸とする糖鎖合成法を組み込んだ新しいスキームに従って高度な機能を持った糖ペプチド分子を作製した。特に、リンパ球と血管内皮細胞との特異的な相互作用(細胞接着)に深く関わる糖鎖であるシアリルルイス X(SLe^x)を同じくインテグリンのリガンド分子であるRGDS含有ペプチドと複合化した新規化合物を合成した。この化合物は同一分子内に2種類のタンパク質(レセプター)を同時に認識する機能を示すので、免疫細胞の機能を制御する際に有効で癌転移阻止剤や抗アレルギー剤などの開発に貢献するリード化合物であることを証明した。この基礎的知見をもとにして創薬のリード化合物を確定し、次年度からこの物質の評価と大量合成に取り組む。
