Research Abstract |
Previous study showed that nuclear binding proteins to vitamin K2 were found in osteoblast. In order to clarify the action of vitamin K2 on osteoblastic function, various concentrations of vitamin K 2 were administered to MC3T3E1 cells, which originated in mouse osteoblasts. Concentrations of 10 7M to 10 5M of vitamin K2 were administered to MC3T3E1 cells not only vitamin K2 alone but also with 10 8M of activated vitamin D. In cell growth, numbers of cells administered various vitamins were higher than that of control cells. Therefore, we investigated the effects サof vitamin K2 on osteocalcin mRNA, receptor activator of NFkB CNK) mRNA, RANK ligand (RANKL) mRNA and osteoprotegerin mRNA. These mRNA levels were increased at 1 hour and 6 after vitamin K2 administration at concentration of 10 6M. Same tendency was observed in addition of vitamin D. Since vitamin K2 seemed to affect cell function, suppression subtractive hybridization method (S SH) were used to detect the factors which might affect cell function. In this method, we detected the genes such as tenascin, lumican, etc. Further investigation is needed to detect the genes which is affected by vitamin K2.
