Research Abstract |
Cancer patients after radiation therapy are given for the effect to survival periods by therapy of an early stage. I attend to factor on the invasion and metastasis of cancer cell and examine to survival periods concerning to there. Recently, the role of U-PA and PAI-1 in malignant cancers has been noticed. Cancer cell invasion into vascular have been considered to make metastasis where the basic membrane and connective tissue are broken down. I use to a specimen that consented to cancer patients, I make to immunostain of U-PA that concerned to the invasion and metastasis and make to immunostain of PAI-1 that action suppressively to invasion and metastasis. To add, I make to immunostain of PCNA that concern to growth. I investigated to use the same factor on blocking degree to culture cell. From 1995 to 1999, concerning to esophageal cancer of patient for the radiation, I make to immunostain for the specimen to get obtain patient. In concern to staining degree, the degree of PAI-1 was 35-55 %, a middle degree. I concluded that this was actioned suppressively to invasion and metastasis. The degree of U-PA was 40-70 %, a comparatively high degree. I concluded that this was advanced to invasion and metastasis. The degree of PCNA was 25-55 %, a comparatively low degree. I conclude that this was a poorly growth. To add, I investigated to blocking degree that I was cultured to the esophageal cancer cell. As a result, cell number was decreased to add PAI-1 for 10, 25, 50 μM. I concluded that this actioned suppressively to cells. And cell number was not change to add U-PA. The cell number was not change to add PCNA for 10, 25 μM, but it was degreased to add 50 μM, After this, I investigated the extracellular matrix for invasion and metastasis of cancer cells and the mechanism for it.
