Research Abstract |
We found that cell cycle related antigen was useful method for cancer diagnosis by Westernblotting. So we investigated whether the method is useful not only for cancer diagnosis but also for sarcoma diagnosis. The materials we used were sera from osteosarcoma, MFH patients and from normal volunteers. These samples were investigated by Western blotting method using proliferative nuclear antigen (PCAN) antibody. Consequently, PCNA positivist was 60 % (3/5) in osteosarcoma, 50 % (2/4) in MFH. In contrast with this, PCNA positivity was 0 % (0/100) in normal volunteers. From this result, we also investigated the usefulness of sandwich ELISA using this PCNA antibody. However, sandwich ELISA method had a problem in lower positivity than in western blotting method caused from low affinity of fixing antibody on the plate. Now we are preparing the other antibody for fixation against PCNA. And we also investigated the mechanism why PCNA appears in sera of cancer patients. We studied the relationship between the number of cell necrosis and PCNA content in the medium. Sarcoma cell line was irradiated 0, 5, 10, 15 Gy for making various necrosis stages. We checked the number of necrotic cells and PCNA content. Consequently, there was high correlation between them. So we thought this was one of mechanism of PCNA appearance in cancer patient.
