Research Abstract |
(1) Fat histomorphometry of beta3 adrenergic receptor agonist (ARa) administered rat models: Ten-week-old female rats underwent sham operation (Sham) or were ovariectimized (OVX) were divided into 4 groups: Sham, Sham+ARa, OVX and OVX+ARa groups. They were given orally the beta3 ARa, BRL 35135 or vehicle daily for 4 weeks and their tibial and lumbar spinal bone and fat histomorphometric parameter were measured. In conclusion, there was closely related between bone remodeling and bone marrow adipocyte numbers, and the beta3 ARa administration and OVX affected the bone marrow adipocyte differentiation, which is site-specific difference between lumbar spine and tibia (Ref 2,5). The beta3 ARa reduced intraabdominal fat without reducing the body weight in oophorectomized obese rats. We also examined the effect of pregnancy and lactation of bone marrow adipocytes in rats and we concluded that the numbers and volume of bone marrow adipocytes suddenly decreased after delivery regardless of lactation or not (Ref 1, 4). (2) Screening of bone mineral density in Shiunji-town: In Sep and Oct of 2000, we measured bone mineral density (BMD) of lumbar spine for 200 women. DNA from peripheral blood samples were obtained with the informed consent of each subject. In the analysis for 7 years, it showed the positive association between the change of calcium intake from dairy food and the change of BMD after 3 years. It also indicated that vitamin D receptor gene polymorphism (Taq I, Apa I) might participate with the association. The gene polymorphism of estrogen receptor, beta AR and uncoupling protein may not participate with it.
