Research Abstract |
We have found the increase of mutation frequency on somatic cells by the analysis for MTH1 gene knockout mouse (KO mouse) by using rpsL gene system. We also found the problem due to the repeat sequences of rpsL gene in this system. The repeat sequences cause sequence slippage at the replication and that directly influence to the mutation frequency. For improve this problem, we have constructed the non-run rpsL gene system and TG mouse by using non-run rpsL gene. We also develop another system using GFP. We introduce the mutation on the GFP and by checking the number of mutated cells that had the reverse mutation. On the other hand, we checked the effect of radiation for MTH1 and MSH2 KO mouse. In MSH2 KO mouse, we recognize the apparent reduction of fertility.
