Research Abstract |
[Materials and Methods] 1. Skin and cartilage-deficit wounds (10x10 mm) were prepared in both ears of a Japanese rabbit. Two wounds were made in each ear. 2. Artificial dermis was attached in two wounds and split skin grafting was performed on the dermis 5,7 and 10 days after attachment (attachment group). In another two wounds, split skin grafting was performed at the same time (control group). 3. The both ears were amputated 7 and 14 days after grafting and these were prepared for samples of molded blood vessels and tissue stained with H-E. Molded blood vessels were viewed under a scanning electron microscope and tissue samples were viewed under a light microscope. [Results] 1. In control group, all grafted skins survivaled completely, on the other hand, there were no survived skins in 5 days after attachment group. In 7 and 10 days after attachment group, grafted skins were survived partially or completely. 2. In control group, H-E stained samples indicated that arrangements of collagen fibers in the grafted bed were irregular as same as those in the scar tissue. On the other hand, the structure of artificial dermis was remained in the grafted bed in attachment group. 3. In control group, centripetal formation of blood vessel plexus in grafted bed, which appeared with wound contraction, was observed. On the other hand, mature formation of blood vessel plexus in the grafted bed was observed in the attachment group. [Conclusion] If secondary skin grafting is performed 7 or 10 days after attachment of an artificial dermis, artificial dermal structure will be remained and it protect the postoperative contraction of the grafted skin.
