動物ミトコンドリアに見出された新規修飾ヌクレオシドの構造機能解析〜ミトコンドリア病発症メカニズムへのアプローチ〜

Research Abstract

我々は、ミトコンドリア(mt)tRNAの点変異に帰属されるミトコンドリア脳筋症(MELAS、MERRF)において、変異mt tRNA^<Leu>(3243位変異と3271位変異)とmt tRNA^<Lys>(8344位変異)は共通にアンチコドン1字目の修飾が欠損していることを見出した(J. Biol. Chem., 275, 4251-4257[2000], FEBS Lett, 467, 175-178[2000])(日医大太田教授との共同研究)。さらに、この修飾の欠損はリボソーム上におけるコドン解読能を低下させ、タンパク合成能を著しく減少させることが発症の根本的な原因であることをつきとめた(EMBO J., 20, 4794-4802[2001])。本プロジェクトでは発症の分子機構を詳細に解析するために、アンチコドン1字目の修飾ヌクレオシドの化学構造を決定することであるが、昨年度すでにその計画をすでに達成した。いずれも置換基にタウリンを含んだ新規の修飾ヌクレオシドで、mt tRNA^<Leu>は5-タウリノメチルウリジン(τm^5U)を持ち、mt tRNALysはその2-チオ置換体である5-タウリノメチル-2チオウリジン(τm^5s^2U)を有することが判明した(投稿中)。タウリンは体内に最も多く含まれるアミノ酸であり、これまで様々な生理活性を持つことが知られているが、タンパク質や核酸など生体高分子の成分としては知られていない。我々はタウリンがこれら新規の修飾ウリジンの直接の前駆体であると考え、有機化学合成した[^<18>O]安定同位体標識タウリンを培地に加えてHeLa細胞を48時間培養した。そこからmt tRNA^<Leu>とmt tRNA^<Lys>を含む5種類のtRNAを単離し、LC/MSにより修飾ウリジンの分子量を測定したところ、いずれも90%以上の効率で培地に加えた[^<18>O]タウリンが取り込まれていることが判明した(投稿中)。この結果はタウリンが生体高分子の成分であることを見出した最初の発見であり、この観点から、様々なタウリンの生理活性を統一的に解釈できるものと考えている。またタウリン欠乏症はmt tRNAの修飾欠損を引き起こしている可能性があり、ミトコンドリア脳筋症との関連から今後の研究をさらに発展していく予定である。
