Research Abstract |
This project has been very important to promote the activity of measure against Echinococcosis in Hokkaido. We have a big problem of serological diagnosis using ELISA, which has been shown unstable data so far. Three kinds of ELISA kit (Hokkaido Institute kit. Em18 kit and Em2 plus kit) are available in the field of serological assay of Echinococcosis. We tried the assay of sera obtained from 863 people living in Sakhalin by using the three kinds of kits. However, we had no correlation between the results obtained by the three kits, Furthermore, we could not diagnose Echinococcosis in 40 ELISA-positive persons by WB analysis and abdominal ultrasonography. The method of ELISA should be reconsidered in the serological assay of Echinococcosis in Hokkaido. In addition, we investigated the echinococcus and the eggs in intestines obtained from 22 foxes caught in Sakhalin Island. No echinococcus and the eggs were detected in those samples. The case study in Sakhalin indicates the important suggestions to our measure against Echinococcosis in Hokkaido. Secondary, we tried to establish the model of mouse echinococcosis by intraperitoneal and subcutaneal infection with protoscoles obtained from echinococcal cysts of peritoneal cavity of the infected cotton rats. C57BL mouse, IFN-gamma KO mouse, nude mouse and SCID mouse were used in the experiments. Among mice, nude mouse seemed to be the best model for the treatment of the secondary echinococcosis. The growing subcutaneal tumor consisted of multiple cysts of echinococcus were observed for 6 months
