Research Abstract	DNA分子をMALDI-TOF式の質量分析法によって測定する場合、その鎖長が長くなるにつれて検出されるシグナルは微弱になり、感度・分解能の低下が引き起こる。このような理由により、長鎖長(100bp以上)のDNAはもとより、比較的短いDNA鎖であっても2塩基鎖の識別はこれまで非常に困難なものであった。本年度はMALDI TOFMSでマイクロサテライト多型識別を可能にするために必要である、長鎖長DNA(100塩基以上)の測定について、2塩基の差長の違いを安定して検出するための至適条件検討を行った。パラメータとしてはまずはじめに(1)精製法、(2)サンプル濃度、(3)レーザー強度に着目し、詳細に条件を検討した。その結果、100bp程度のDNA鎖について、2塩基差の識別に十分な分解能を得ることが可能となった。また、分子量の高い3〜5塩基繰り返しマイクロサテライトについても、チップ上で質量分析法を用いた分子量測定によって多型性の検索が可能となるように、測定条件を検討している。今年度は、赤外線レーザーによる照射が可能な測定装置の開発を行った。現在、これら諸条件のさらなる検討により、マイクロサテライト多型検索のための質量分析によるDNAチップ法の能力向上に取り組んでいる。
