Research Abstract

IgM抗体は、病原微生物の初回感染後にBリンパ球から分泌される免疫グロブリンである。またIgM抗体は、自然抗体として病原体に感作されていない個体にも存在することから、生体防御反応の初期段階に重要な役割を担っていることが考えられている。しかし、IgM抗体と結合した病原体の排除機構や獲得免疫系における役割はまったく明らかにされていない。申請者はIgM抗体に対するヒトおよびマウスのFc受容体(Fcα/μレセプター)を世界に先駆けて同定した。さらに申請者は、Fcα/μレセプターがB細胞やマクロファージなどの抗原提示細胞上に発現しており、これが黄色ブドウ球菌などの病原微生物とIgM抗体との免疫複合体の抗原提示細胞内への取り込みに直接関与していることを発見し、抗原提示細胞による新しい病原体取り込み経路の存在を証明した。本研究では、Fcα/μレセプターのノックアウトマウスを作製し、Fcα/μレセプターを介して取り込まれた病原微生物が抗原としてT細胞に提示され、獲得免疫系の作動に関与していることを証明する。
本年度はFcα/μレセプターのノックアウトマウスの作製を試み、マウスFcα/μレセプターの染色体遺伝子のクローニングとその構造解析を行った。これをもとに、さらにターゲッテイングベクターの構築をおこない、組み換え体ES細胞を得た。
