Research Abstract

本領域は総括班と4つの計画研究班及び公募研究から構成され、緊密な連絡の下に領域全体および各班の研究目標を達成するように推進された。各研究班の研究成果の概要は以下の通りである。
(総括班)1)日本RNA学会と共催で国際シンポジウム「The New Frontier of RNA Science」を開催した(平成15年11月24-27日、於京都国際会議場)。国外招待講演者25名を含む計380名の参加者のもとに学術的、教育的、国際交流的に実り豊かな第一級の国際会議となった。2)計画研究項目の内容に関して最新の調査点検議論を実施した。3)合同班会議を平成16年2月2〜4日に小樽市マリンヒルホテル小樽において開催した。4)第4回総括班会議を平成15年11月23日(於京都市)に開催した。5)RNA Network Newsletter第3、4号を発行し、メディアを含む関係方面へ各2000部配送し、領域研究の啓蒙を行った。
(A01班・RNPマシーン)酵母2型解離因子eRF3の結晶構造の解明、ペプチド解離の触媒塩基の発見、再生因子RRFのEF-Gの転移活性非依存性等の証明等、リボソーム研究を推進した。又、古細菌と真核生物の各種アミノアシルtRNA合成酵素の単体や共結晶の構造解明tRNA修飾酵素の結晶構造と反応機作の解明、ミトコンドリアtRNA修飾異常による疾患発症の分子基盤解明を行った。
(A02班・RNA制御スイッチ)翻訳伸長と解離のスイッチを担うリボソームRNA塩基の同定と活性中心変異体の系統的な分離解析により動的作動機構に関する知見の集積、植物における翻訳と共役したmRNA分解制御、精巣特異的細胞質ポリ(A)ポリメラーゼの欠損マウスの作製と解析、RNA関与MAPK信号伝達系のフィードバック制御の発見等、RNA制御スイッチに関する重要な知見をえた。
(A03班・動くRNA)神経特異的RNA結合蛋白質Huの線虫ホモログによる老廃物排出器官形成制御の発見、スプライシング調節RNA結合タンパク質Y14による線虫の生殖細胞の性分化制御の発見等、スプライシングやmRNA核外輸送のカップリング等に関する分子基盤が明らかにされた。
(A04班・高次複合系RNA動態)RNA干渉(RNAi)は真核生物の遺伝子制御の新パラダイムであるが、脆弱X症候群原因遺伝子(FMR1)のショウジョウバエ相同体の解析により「RNAi関連分子装置の異常による疾患」というヒト分子遺伝学における全く新しい領域を開いた。
