制御性T細胞による免疫自己寛容維持機構の解析とその機能操作による自己免疫の修復

Research Abstract

制御性T細胞の発生・分化および機能の分子的基礎について、昨年度に引き続き研究を進め以下の研究結果を得た。
(1)本年度、制御性T細胞と活性化T細胞を区別できる単クローン抗体の作製を試みた。マウス制御性T細胞に対してそのような特異性を持つ抗体TH6を得た。CD25+CD4+制御性T細胞は、一般の活性化T細胞よりもTH6分子を高発現する。従って、TH6の投与により、マウスの進行癌に対し有効な腫瘍免疫応答を惹起できた。また、アロ移植抗原の刺激により活性化したT細胞群より、TH6高発現T細胞を調整すると、それらは、移植片の拒絶を有意に抑制した。一方、正常マウスにTH6抗体を投与すると自己免疫病を惹起できた。この結果から、病的、生理的免疫応答において、エフェクターT細胞のみならず、制御性T細胞も抗原に感作され、抗原特異的に増殖し、抗原特異的抑制を示すと考えられる。また、TH6分子の操作による免疫応答の制御の可能性を示す。
(2)同様に、ヒトの制御性T細胞に特異的分子を、マイクロアレイ、単クローン抗体の作製など様々な方法で探索を進めた。いくつかの単クローン抗体を得、解析を進めている。
(3)制御性T細胞に高発現するGITR(Glucocorticoid-induced TNF receptor family-related gene)のリガンドであるGITR-Lについて、その機能を解析した。興味深いことに、GITR-Lを介するGITR刺激は、制御性T細胞の抑制能を解除した。この性質を利用して、GITR-Lをレトロウイルスベクターにより腫瘍に発現させたところ、有効な腫瘍免疫応答を惹起できた。
