制御性T細胞による免疫自己寛容維持機構の解析とその機能操作による自己免疫の修復

Research Abstract

自己に対する免疫不応答(免疫自己寛容)の導入・維持機構の解明は、自己免疫病の予防・治療、自己から発生した腫瘍細胞に対する効果的な免疫応答の惹起、さらには、移植臓器の長期生着をはかる上で重要である。本研究では、制御性T細胞による免疫制御機構の分子的基礎を解析し、自己免疫、腫瘍免疫、移植免疫への応用を目指す。本年度は、制御性T細胞による抑制の分子機構、および転写因子Foxp3が制御性T細胞の発生、機能に果たす役割を解析した。(1)前年度に引き続き、DNAマイクロアレイを用いた場合に制御性T細胞に高発現する遺伝子についてその機能を解析した結果、制御性T細胞に極めて特異性の高い遺伝子として、Gpr83,Ecm1,heliosを同定した。前2者は、Foxp3遺伝子の支配下にあり、heliosは、Foxp3によって制御されない。これらの分子に対する単クローン抗体の作製を試みた。(2)GITR-L分子の制御性T細胞活性化における役割について、GITR-L欠損マウスを作製し、純系マウスに戻し交配した。その結果、一部のマウスは自己免疫病を自然発症した。その発症機構について解析を進めた。(3)制御性T細胞の発生および機能のマススター制御遺伝子であるFoxp3について、Foxp3-GFPノックインマウスを作製するべく、現在キメラマウスの段階まで作業を進めた。(4)Foxp3遺伝子の発現制御について、Foxp3分子と結合する分子を、yeast two-hybrid法にて解析を進め、6個の特異的に会合する分子を同定した。現在、それぞれについて解析を進めている。
