Research Abstract |
癌の骨転移は悪性黒色腫B16マウスメラノーマ細胞を用いて、骨髄に対する接着並びにこれに伴う骨の破壊についての検討を行った。特に長管骨の骨幹端における骨髄内の癌の転移並びに細胞の接着を骨髄内への直接の癌細胞の埋植によって検討した。実験にはオステオポンチンノックアウトマウス並びにその野生型の対照となるマウスを用い、大腿骨の骨髄に対して、0.6mmのKワイヤーを挿入し骨髄を完全に除去した後、この空間に対する癌細胞の播種を行った。動物は癌細胞の播種の後に経時的にレントゲン撮影を行いその結果、悪性黒色腫細胞による骨の破壊が観察された。オステオポンチンノックアウトマウスと野生型のマウスの比較検討によりノックアウトマウスにおける骨の吸収窩の数の低下が明らかとなった。癌細胞の骨髄内埋植後、4週間の時点において動物を屠殺し、マイクロCTによって骨の破壊レベルを検討した結果、骨髄内における海綿骨の破壊と皮質骨の破壊が野生型では時間と共に亢進したのに対しオステオポンチンノックアウトマウスでは骨髄内の海綿骨並びに皮質骨は悪性黒色腫の播種後にもかかわらず構造が保たれていた。組織学的にはこれらの細胞の周辺における破骨細胞の形成とこれによる骨破壊が野生型では著明に観察されたのに対しノックアウトマウスにおいては破骨細胞の数、ならびにそれによって起こる吸収窩の形成は抑制されていた。以上のことから骨髄に対する直接の悪性黒色腫細胞の播種実験によりオステオポンチンの存在が骨の癌転移巣における破骨細胞の誘導に促進的に関与すること、また破骨細胞自身による骨の破壊にも寄与することが明らかとなった。
