Research Abstract	ABO式血液型遺伝子の上流域の解析から、1)転写開始点の上流2kb〜0.9kbにAlu、LINE領域があり、それらの領域はメチル化されている。2)転写開始点の上流0.7kbに新たな転写開始点を同定したが、その周囲には細胞特異的な発現を示すプロモーターが存在する。3)新たに同定された転写開始点はCpG islandの上流側の辺縁にあたり、そこから下流約1.3kbの範囲がCpG islandになっている。4)以前に同定された転写開始点の周囲には細胞非特異的なプロモーターが存在する。5)癌細胞ではABO式血液型遺伝子の上流域のDNAメチル化はAlu、LINE領域から拡大して、下流の転写開始点に及ぶことがあった。 H遺伝子の上流域の解析から、1)FUT1遺伝子の転写開始点が少なくとも3カ所に存在する。2)それら3つの転写開始点が組織特異的或いは細胞の分化度に特異的に利用されている可能性を示した。3)大腸癌においてエキソン1及びエキソン7の転写開始点が利用されている事を5'-RACE法を用いて同定した。4)さらに未分化な大腸癌ほど上流のエキソン1が転写開始点として利用される。5)大腸癌をSodium butyrate処理により分化誘導すると転写開始点がエキソン1からエキソン7に移行する事を見いだし、エキソン1の上流及びエキソン7の上流約1kbの領域をレポーター遺伝子の上流に組み込みその転写活性を測定したところ大腸癌において転写活性が存在する事が示された。
