| Research Abstract |
1.グルココルチコイド(GC)刺激による胸腺綱胞のアポトーシスに先行して発現誘導されるTDAG8の機能につき検討した。
2.胸腺細胞は、放射線照射、GC刺激、抗CD3抗CD28刺激により、アポトーシスにおちいる。TDAG8は、GC刺激によるアポトーシスに先行して強発現するが、放射線照射によるアポトーシスでは、発現しない。一方、抗CD3抗CD28刺激においては、GC刺激に比して弱いながら発現が認められた。したがって、TDAG8は、GC刺激によって、誘導されるアポトーシスに特異的に関与する分子であることが示唆された。
3.TDAG8遺伝子導入マウスを樹立した。無刺激の状態でも、胸腺は萎縮しており、CD4^+抗CD8^+胸腺細胞の著名な減少を認めた。遺伝子導入マウスでは、GCのin vivo投与により、胸腺の皮質細胞のアポトーシスが著明に増強した。
4.TDAG8遺伝子導入マウスでは、GC投与により、カスペース3,8,及び9の活性化が著名に亢進した。
