Research Abstract |
Results outline of the research: 1. Mouse embryonic stem cells (ES cell) were cultured with several growth factors and expression of transcription factors and hepatocyte specific markers, such as albumin and alfa-phetoprotein (AFP) were confirmed with RT-PCR after 3 weeks of treatment, showing that murine, embryonic stem cells could be differentiated to hepatic lineage. 2. A new differentiation system, co-culture of mouse ES cells and mouse hepatocyte, has developed and demonstrated the expression of albumin and AFP after 2 weeks, suggest that this system may also useful to differentiate ES cells to hepatic lineage. 3. Monkey ES cell were treated with aFGF for 3 weeks, and expression albumin and AF P mRNA were verified with RT-PCR. 4. A new cell-tracking technology, visualization of labeled cells under magnetic resonance imaging (MRI) in living animals was established. This new method is applied to the animal model of cell transplantation to mice which had partial hepatectomy. Mouse hepatocyte like cells derived from ES cells, which were cultured with HGF, FGF and EGF, were labeled with Fe, and were transplanted into spleen of nude mouse with partial hepatectomy. Labeled transplanted cells were able to be traced with MRI with time. This new method would be strong tool to in the field of stem cell and cell transplantation research.
