Research Abstract	本年度は、初年度に引き続き、着床を標的にした妊孕能の制御可能なモデルマウスの開発を目指すべく、子宮への外的・物理的遺伝子操作によるシステムの確立を試みると同時に、誘導プロモーター下Cre-loxPシステムを用いた遺伝子改変マウスを作成した。 1.子宮腔へのcalcium bindidng protein D-9K(Cabp9K)遺伝子およびantisense oligodeoxynucleotide(antisense ODN)導入システムの確立着床期特異的発現パターンを示すことが報告されているCabp9Kを解析対象遺伝子として、その遺伝子導入のための条件設定を行うと同時に、Cabp9K発現ベクターおよび蛍光標識antisense ODNの作成とそれらの機能検定を行い、マウス線維芽細胞株を用いたin vitro遺伝子導入システムにおいて、それらが機能しうることを確認した。一方、LacZあるいはGFP発現ベクターなどをレポーターとして、HVJ-envelopeや様々なliposome系の導入試薬を用いて、子宮への導入遺伝子の浸透度・分布度の解析を行った。既報に比べて低い導入効率しか得られず、既報の方法の再検証の必要性を示唆する新しい知見と、今後のシステム開発のための有用な基盤情報が得られた。 2.Cre-loxPシステムを用いた妊孕能制御可能マウスモデルの作成の試みとその検定子宮内膜特異的に発現する遺伝子のプロモーターによる複数系統のCre発現トランスジェニックマウスを作製した。Germ line transmissionを確認すると同時に、in vitroおよびin vivoにおいて子宮特異的にCreを誘導し得ることを、RT-PCRおよび免疫組織染色により確認した。
