Research Abstract |
This study was conducted in an attempt to search endocrin disrupting chemicals, using male chickens or cultured hepatocytes of male chickens. The vitellogenin mRNA expression was selected as an index. Vitellogenin is the egg yolk protein expressed in the liver of female chickens. The chemicals with estrogen-like effect induce the vitellogenin expression in the liver of male chickens. Vittellogenin mRNA was detected using the rt-PCR methods. Using this method, we found the estrogen-like effect of an insecticide (Name of the chemical is not shown. ). Furthermore, we investigeted the pollution by endocrin disrupting chemicals of the water of Lke Kasumigaura, using male chickens. The vitellogenin mRNA expression could not be detected in the liver of male chickens administered the lake water. A primary culture of hepatocytes was applied to experimental models of liver function. In this study, cultured hepatocytes with non-serum medium were used to search endcrin disrupting chemicals. Vitellogenin mRNA expression was detected in cultured hepatocytes with or without estrogen. This noize could be eliminate within this project term.
