| Research Abstract |
TTウイルス(TTV)は多様性に富むウイルスであり、数多くの遺伝子型が認められる。これまで、1型から29型までの少なくとも29種類同定されているなかで1型のTTVに肝障害との関連が示唆されている。この関連を明らかにしていく上で遺伝子型に特異的かつ簡便に検出できる抗原抗体測定系の確立が必要である。本報告は、先ず1型の、特にプロトタイプである1a型に特異的な抗原抗体検出系の確立を目的として行った研究実績の概要を記す。
TTVの遺伝子型1aのORF1〜ORF4蛋白質に特異的な抗原抗体測定系を構築するために、(1)抗原とする蛋白質発現ベクターの構築、(2)蛋白質の発現と精製、(3)抗原の免疫と抗体の作製、の手順で行った。
1.TTV ORF1〜ORF4蛋白質の発現ベクターの構築
ORF1〜ORF4蛋白質の発現を大腸菌およびバキュロウイルスを用いた昆虫あるいは昆虫細胞系を用いて行うために、夫々に適したベクターに各発現遺伝子を改変し挿入した。ORF1蛋白質については数種の断片化した遺伝子を作製し、ORF3とORF4蛋白質についてはイントロンを削除した遺伝子を作製した。
2.発現と精製
大腸菌による発現では、宿主株、培養温度および発現誘導条件(時期、誘導剤濃度、培地)等を検討し、最適化を行った。バキュロウイルスを用いた発現では、カイコ幼虫での発現を行った。
3.抗原の免疫とモノクローナル抗体の作製
精製した蛋白質をマウスに免疫注射してポリクローナル抗体を取得し、現在モノクローナル抗体の作製を行っている。
