口腔癌細胞のspectral karyotyping法による染色体構造異常の解析

| Research Abstract |
Spectral Karyotyping(SKY)法用いてヒト扁平上皮癌細胞の染色体構造異常の解析を行った。また染色体標本の作製について染色体作成法とSKY paint kitの適切なハイブリダイゼーションの条件について再検討した。0.075M KCLと0.9%クエン酸溶液の混合比を3:1にすることで良好な染色体の伸展ができ、メタメタフェーズスプレッダー用機器のHANABIを使用することで染色体が重ならずに展開でき、多数のメタフェーズ細胞をプローブを落下する領域に展開できるようになった。またManufacture's protocolに先立ち、2×SSC処理を行うと、良好名ハイブリダイゼーションすることを見いだした。
以前の実験に対して新たに9腫の口腔扁平上皮癌細胞(KM-2,Sa-3,H-1,TYS,BHY,HN,OSC19,OSC70,SAS)について検討した。染色体構造異常と著明な変化は9個の細胞中8個の細胞で5番染色体のisochromosomeが認められた。
SKYで確認された5pおよび8qのisochromosomeはFISH法により証明した。さらに、それらはCGH法で検出される5Pや8qのwhole arm gainと密接に関わっていた。これより口腔扁平上皮癌は同領域のコピー数の増加が高頻度にみられるが、それらに加えてisochromosomeの形成が関わっているものと推察された。
