Research Abstract	HIVウイルスに対する生体内の免疫応答をリアルタイムでとらえ、そのT細胞レセプターの遺伝子情報を獲得し、効率良くリンパ球に遺伝子導入する技術を中心とした、ウイルス特異的T細胞再構築システムの確立を目的とした研究を進めた。マウスのHIV免疫応答モデルにおいて、CTLのエピトープと判明しているP18IIIBペプチドを樹状細胞にパルスし、これでBALB/cマウスを免疫した。さらにこの集団で細胞傷害性T細胞に主に使われているT細胞レセプターのV領域の分析から、Vα2とVβ7を選択した。次にこの両者が陽性の1つの細胞に着目し、免疫染色とセルソーターによる単一細胞分離法とRT-PCR法により、そこに発現しているT細胞レセプターのα鎖、β鎖をクローニングした。これらを別々のレトロウイルスベクターにクローニングし、パッケイジング細胞に導入後、脾臓細胞に遺伝子導入したところ、HIVエピトープを発現している細胞に対して細胞傷害活性を示すことが明らかとなった。さらに、生体内での機能を探索する目的で、上記エピトープをパルスした脾臓細胞をCFSEで標識し、これを標的とした生体内細胞傷害試験をおこなったところ、実際の細胞傷害を確認した。以上の結果から、我々が目指しているT細胞レセプターの試験管内機能再構築が基本的に可能であることが示された。
