体性幹細胞の心筋細胞への特異的分化誘導

Research Abstract

骨髄間葉系細胞は、様々な間葉系細胞を作る幹細胞を有している。幹細胞からは様々な細胞が分化可能であり、臨床応用へは特異的分化誘導法の確立が必要である。また、不全臓器の機能改善をもたらすだけの細胞数が必要である。しかしながら、ヒトの細胞はげっ歯類の細胞とはことなり極めて増殖能が乏しいことが大きなハードルである。
本研究では、特異的分化法の確立と幹細胞としての性質を保持しながらの増殖誘導を目標に、遺伝子導入によるCommitment/Proliferationのコントロールが可能であるか検討した。心筋細胞への初期分化に必須なNkx2.5とGATA4を間葉系幹細胞に導入することで、分化高率はそれぞれの導入により1オーダー上昇し、両方の導入により2オーダーの効率上昇をin vitroにおいて示した。しかしながら、これをin vitroに移しても、心筋細胞への分化効率はin vitroでの効果は認めることが出来ず、遺伝子導入をしていない細胞の移植と同等の分化しか示さなかった。この結果は、ドナー細胞のin vivo環境への生着が大きく影響しており、移植から生着に至るまでに、最近の報告から考えると少なくとも2オーダーの数の上での減少があることが、病理学的評価を困難なものにしていると考えている。
一方で、ヒト骨髄由来間葉系細胞にTelomerase/E6/E7/Bmiを遺伝子導入することで、Proliferation doublingの延長を認めた。また、これらの遺伝子導入細胞は、心筋細胞を含む骨・軟骨・脂肪といった間葉系細胞への分化能を保持していた。この細胞を心臓へ移植する実験において、心筋細胞への分化能が保持されていることが示された。
