Research Abstract |
In order to clarify the function of two kinds of histone H1 variants (meiosis-specific histone mH1, male gametic nucleus-highly concentrated histone gH1) that had been detected during male gametogenesis in Lilium longiflorum, we tried to produce transgenic tobacco which were introduced the cDNA of mH1 by Agrobacterium-mediated system. We also produced transgenic tobacco introduced the cDNA of gH1 with GFP (green fluorescent protein) as a reporter gene. As a result, we obtained several transgenic tobacco plants which was biochemically proved to express the foreign mH1 besides tobacco histone H1 and whose somatic nuclei constantly fluoresced green light. They normally developed and formed flower buds, though they contained lily linker histone in the chromatin. The pollen was fertile. Immunoelectron microscopical observation demonstrated that gH1 was concentrated in the heterochromatin areas. Therefore, it was suggested that gH1 was related to the formation of heterochromatin as in the case of generative nucleus, but had little effects on the gene expression. In this study, although the planed experiments were successfully achieved, we could not clarify the function of lily histone H1 variants. However, because we obtained unique transgenic tobacco, we will be able to observe nuclear division successively and to analyze he process during nuclear fusion after cell fusion in the future.
