Research Abstract |
Tow alkaline phosphatase genes, Alp-m and alp-s, were independently cloned from the silkworm, Bombyx mori. They were mapped to a small DNA region and shown to be in tandem. Exon-intron structures of the two genes were highly conservative. The similarity of the nucleotide sequences was significantly high (60-79%) in the exons of result in four types of the intervening sequences and deletion of the Alps gene result in four types of polymorphic structure in the Alp cluster. The similar structure of the cluster was found in the genome of Bombyx mandarina, one of the evolutionally closest species to B. mori, but in other species of wild silkmoth. These results suggest that Alp-gene cluster was originated by a duplication of their common ancestor gene before the Speciation of B. mori and B. mandarina. Because many copies of different transposable genetic elements existed in and around the Alp genes in Bombyx, we examined P and hobo elements of Drosophila melanogaster for understanding their mechanism of invasion, propagation, and inactivation, in relation to their host genomes.
