| Research Abstract |
表皮角化細胞が分化の過程で発現する各種タンパクの相互作用を解析した。本年度の主な研究成果は以下のごとくである。
1.分化型ケラチンのひとつであるK1のテールドメインに変異が導入されることによって生じる角化異常症であるCurth-Macklin型豪猪皮状魚鱗癬では、他の研究者の報告ではそのような異常なケラチンは異常に核内移行すると報告されていたが、これは少なくともわれわれのin vivoの系では認められないことを報告した(J Invest Dematol 2003)。また、他の研究者が指摘していた異常ケラチンによるデスモゾームの障害も認められないことを報告した(J Invest Dermatol 2003)。
2.角化細胞が分化に伴って合成する顆粒である層板顆粒の実態について報告した(J Invest Dermatol 2004)。この顆粒は従来考えられていたような孤立性の顆粒ではなく、ゴルジ複合体の延長構造として網目状に細胞内に発達している構造であることが判明した。また、各種分泌酵素、構造タンパク、セラミド前駆対がこの構造の中に含まれ、独立して輸送されることも示した。
