Research Abstract |
chromosome 10q has been reported for its deletion especially in glioblastoma multiforme. The locus of FGFR2 gene is on chromosome 10q25.3-26,this region is often deleted especially in glioblastoma multiforme. We investigated the presence of FGFR2 gene by means of Southern blot and it revealed FGFR2 gene exists. Loss of heterozygosity(LOH) is reported to occur in this chromosomal region, therefore we investigated the possibility of FGFR2 variant. To see the transcription of the incomplete mRNA by LOH, one-direction PCR was performed using the PCR primer specific to the known exon of FGFR2. However there has been no PCR product detected. Further analysis is now proceeding to find any variant. Gene silencing is another mechanism for no mRNA nevertheless the gene exists. The methylation of the promoter was analyzed and revealed its hypermethylated promoter. This is thought to be one of the mechanism of gene silencing of FGFR2. We are constructing the known FGFR2-variant vector what is supposed to be introduced into glioblastoma cell lines. The growth, morphology and mobility will be analyzed. Supposed that FGFR2 has tumor suppressing effect, the way of induction of FGFR2 in malignant gtioma must be found. The real function of FGFR2 is still unknown. Dimerization of FGFR1 and FGFR2 is gives so-called dominan-negative effect may be the core of tumor suppressive effect in glial tumors.
