マキシKチャネルの細胞内機能因子による新しい活性化調節機構と生理的役割の検討

Research Abstract

1.平滑筋におけるマキシKチャネルの役割について、収縮機能実験・電気生理実験・遺伝子発現実験・ノックアウト動物を用いて多角的な検討を行い、以下の新知見を得た。
2.細胞内cAMP濃度上昇とマキシKチャネル活性化という観点から血管平滑筋の弛緩反応を解析したところ、IPアゴニスト以外に、β-アドレナリン受容体活性化薬(イソプレナリン)の弛緩作用に細胞内cAMP濃度上昇を介さないマキシKチャネル活性化機構が有意に寄与する可能性を見出した。
3.イソプレナリンの気管平滑筋弛緩作用がマキシKチャネルの選択的阻害薬であるイベリオトキシンにより強力に抑制されることを見出し、β_2-アドレナリン受容体-マキシKチャネル活性化連関が生理活性物質や喘息治療薬の気管(支)平滑筋の弛緩作用の主経路である可能性が強く示唆された。尚、マキシKチャネルの活性化におけるcAMPの役割については現在検討中であり、血管平滑筋との差異が今後の研究の焦点である。
4.膀胱平滑筋の自発性筋原収縮と活動電位の発生がイベリオトキシンにより著明に増強されることを見出し、この平滑筋では、非刺激時の筋の緊張性制御においてもマキシKチャネルが重要な役割を果たしている可能性が示された。また、マキシKチヤネルのβ(β_1)-サブユニットを活性化するエストラジオール・タモキシフェンに筋原収縮と活動電位の抑制効果を新たに見出した。
5.β_1-サブユニット欠損マウスの大動脈平滑筋では、wild-typeと比較すると、細胞外カリウム濃度昇、BayK8644(カルシウムアゴニスト)に対する感受性が著しく亢進していることを見出した。
6.以上の結果から、(1)平滑筋の緊張性制御機構におけるβ-アドレナリシ受容体-マキシKチャネル活性化連関の生理的意義と、(2)マキシKチャネル活性化調節におけるβ_1-サブユニットの重要性が明らかとなった。
