Research Abstract |
Type 2 iodothyronine deiodinase, which converts T4 to T3, is mainly expressed in central nervous system and anterior pituitary in the rat. We have demonstrated strong expression of type 2 iodothyronine deiodinase in human thyroid gland. We have also reported that patients with autoimmune thyroid disease have antipituitary antibodies. Based on these results, we have speculated that patients with autoimmune thyroid disease may have antibodies against type 2 iodothyronine deiodinase which is highly expressed in both human anterior pituitary gland and thyroid gland. We measured antibodies against type 2 iodothyronine deiodinase by using iodothyronine deiodinase activity assay or synthetic peptide corresponding to the active center of type 2 iodothyronine deiodinase. We have found positive type 2 iodothyronine deiodinase antibodies in patients with autoimmune thyroid disease, such as Graves' disease and Hashimoto's disease. Type 2 iodothyronine deiodinase antibodies are positively correlated with antipituitary antibodies determined by ELISA using rat anterior pituitary as an antigen. We have also studied the expression and the regulation of type 2 iodothyronine deiodinase in mouse pituitary tumor cells, which has been used for detecting antipituitary antibodies. These results suggest that type 2 iodothyronine deiodinase is a candidate for an antigen recognized by antipituitary antibodies.
