Research Abstract |
磁場には、磁石や地球地磁気などによる定常磁場と、交流電流から発生する変動磁場の大きく二種類が存在するが、変動磁場については携帯電話の普及などによって磁揚に曝される機会が増えるにつれて、その悪影響が近年懸念されている。これに対して、磁場曝露の疾患治療効果を期待させる報告例も少なくない。例えば、精神科領域では長年電気けいれん療法が行われてきたが、最近ではより安全でかつ簡便な非侵襲的代替療法として、磁場を応用した反復性経頭蓋磁気刺激法の有効性が、多くの臨床的研究で示されている。特に、難治性うつ病、強迫性障害、あるいは統合失調症(精神分裂病)等の患者で、磁気刺激による症状改善例報告があることは興味深い。今回は、初代培養神経細胞の神経活動に対する定常磁場曝露の影響について検討した。ラット胎仔脳より調製した海馬由来初代培養神経細胞は、培養日数の増加とともに形態と機能を変化させて、細胞体から1本の軸索と複数の樹状突起の伸長と極性を発達させたのちに、神経細胞間でのシナプスネットワークを形成して成熟した。この海馬由来神経細胞を、調製後3時間目より磁束密度100ミリテスラの磁場環境下において、無血清条件下に培養すると、細胞生存率には影響は見られなかったが、磁場曝露による神経細胞マーカー蛋白質MAP2発現の著明な低下とともに、アストログリア細胞マーカー蛋白質であるGFAP発現量の増加が誘発された。MAP2は、脳内においては神経細胞に特異的に局在し、微小管の安定あるいは樹状突起の伸展に不可欠である。したがって、持続的な定常磁場曝露は細胞障害を誘発せずに、海馬神経細胞において神経細胞の成熟度に強い影響を与える可能性が示唆される。
